SCA® Motility and concentration is a program for the automatic analysis of concentration and motility parameters of sperm.
The software detects the motile/immotile spermatozoa automatically, performs an accurate count and accordingly provides the concentration results. At the same time, motile sperm analyses is performed by following the sperm trajectory and determines the velocity. The results are then presented within seconds taking into account the W.H.O. motility parameters (both percentage motility ratings as well as the a to d WHO ratings): VCL, VAP, VSL, STR, LIN, WOB, ALH, BCF.
According to W.H.O. analysis of 200 sperm is required, which is approximately equivalent to 2 or 3 fields to be captured. Analysis time is one second per field.
SCA® has Full and Lite versions for motility and concentration.

Motility parameters
VCL: Curvilinear velocity (µm/s). Time/average velocity of a sperm head along its actual curvilinear trajectory, as perceived in two dimensions under the microscope.
VAP: Average path velocity (µm/s). Time/average of sperm head along its spatial average trajectory. This trajectory is computed by smoothing the actual trajectory according to algorithms in the CASA instruments these algorithms vary among instruments.
VSL: straight/line velocity (µm/s), Time/average velocity of a sperm head along the straight line between its first detected position and its last position.
ALH: Amplitude of lateral head displacement (µm). Magnitude of lateral displacement of a sperm head about its spatial average trajectory. It can be expressed as a maximum or an average of such displacements. Different CASA instruments compute ALH using different algorithms, so that values are not strictly comparable.
STR: Straightness. Linearity of the spatial average path, VSL/VAP.
LIN: Linearity. The linearity of the curvilinear trajectory,VSL/VCL.
WOB: Wobble. Measure of oscillation of actual trajectory about its spatial average path.
BCF: Beat/cross frequency (beats/s). The time-average rate at which the curvilinear sperm trajectory crosses its average path trajectory.
Following the W.H.O. suggestions, (WHO laboratory manual for the examination of human semen and the sperm-cervical mucus interaction, 1999) is recomendable to analize at least 200 sperms for sample. With the CASA SCA it means capturing an average of 2-3 fields for sample. One second for field is the analysis time.
The SCA is available in two versions: the FULL version For Motility & Concentration and the LITE version.
There also the option to ampliate the LITE version to a FULL version in a second time.
