caus\u00e9es par l\u2019existence d\u2019un num\u00e9ro et \/ ou de la structure anormaux des chromosomes.<\/p>\n<p>Les chromosomes sont des structures complexes qui se trouvent dans le noyau de la cellule. Ils sont compos\u00e9s d&rsquo;ADN, \u00e9troitement enroul\u00e9s avec des prot\u00e9ines appel\u00e9es histones qui soutiennent la structure, prot\u00e9ines de non histones, ARN, et les polysaccharides.<\/p>\n<p>Durant le processus de la mitose, les 23 paires de chromosomes humains se condensent suffisamment pour \u00eatre visibles sous microscope en champ clair avec l&rsquo;objectif 100x (huile). Les chromosomes sont expos\u00e9s \u00e0 un inhibiteur mitotique qui bloque la formation de l\u2019appareil microtubulaire et arr\u00eate la division des cellules au stade de la m\u00e9taphase.<\/p>\n<p>Il peut \u00eatre utilis\u00e9 plusieurs types de tissus pour la pr\u00e9paration des chromosomes. Par exemple: le sang p\u00e9riph\u00e9rique, la moelle osseuse, le liquide amniotique, et autres produits de conception. Le colorant colore les r\u00e9gions des chromosomes riches en Ad\u00e9nine (A) et Thymine (T), ce qui produit une bande sombre.<\/p>\n<p>Hybridation Fluorescente In Situ (FISH ) est une technologie utilisant des sondes d&rsquo;ADN marqu\u00e9es par fluorescence pour d\u00e9tecter ou confirmer des g\u00e8nes ou anomalies chromosomiques . L&rsquo;ADN de l&rsquo;\u00e9chantillon (des chromosomes en m\u00e9taphase ou des noyaux en interphase ) est d&rsquo;abord d\u00e9natur\u00e9 , un processus qui s\u00e9pare les brins\/chaines compl\u00e9mentaires au sein de la structure en double h\u00e9lice de l&rsquo;ADN .<\/p>\n<p>La sonde de fluorescence \u00a0est alors ajout\u00e9e au m\u00e9lange de l&rsquo;\u00e9chantillon d\u00e9natur\u00e9 qui s\u2019hybride avec l&rsquo;\u00e9chantillon d&rsquo;ADN au niveau de la zone cibl\u00e9e. Le signal de la sonde peut alors \u00eatre vu par microscopie de fluorescence.
